Question: Maneka Mirchandaney - Evercore ISI - Analyst
: To start, I guess just give us a quick snapshot of where Kymera is right now.
Question: Maneka Mirchandaney - Evercore ISI - Analyst
: Awesome. Yes. Let's actually just start with the R&D Day, with that coming up in a couple of weeks. I know you guys have said the MAD healthy
volunteer update will be there but -- for IRAK4 -- but what else should we be focused on in that presentation?
Question: Maneka Mirchandaney - Evercore ISI - Analyst
: Awesome. Well, looking forward to that for sure. But want to start with 474 on -- and I think a lot of us have seen the SAD update that you guys
presented a few months ago. You also presented the cytokine ex vivo data at TPD. Maybe just help us contextualize that data. Like, what does that
ex vivo cytokine reduction mean, and how translatable do you think it is to the in vivo setting as well?
Question: Maneka Mirchandaney - Evercore ISI - Analyst
: Got it. That makes sense. And in the MAD update, I think, in addition to the circulating cells, you guys are also looking at skin biopsies. From the
preclinical data, it looks like they correlate pretty well. But just give us a sense for your confidence in seeing that in the clinic as well.
Question: Maneka Mirchandaney - Evercore ISI - Analyst
: Got it. That makes sense. In the SAD data at the high doses, it looks like you get suppression of IRAK4 out multiple days after dosing. How do you
think about different dosing frequencies as a potential option for this therapy beyond daily, if you think you could kind of push dosing out less
frequently, or why not?
Question: Maneka Mirchandaney - Evercore ISI - Analyst
: As we think about moving from the healthy volunteer setting this year to patients next year, are there any differences in expression of IRAK4 levels
or anything else that might impact the doses that you think you might need between healthy volunteers and patients?
Question: Maneka Mirchandaney - Evercore ISI - Analyst
: That makes sense. And in that patient data update in the middle of next year -- beyond degradation, what should we be focused on in those
patients? And would you expect any early signs of disease modification, and how do you think this would manifest in terms of endpoints?
Question: Maneka Mirchandaney - Evercore ISI - Analyst
: Yes, that makes sense. I want shift gears to some of the pipeline programs. Starting actually with the STAT3 degrader, we saw some preclinical data
on that recently at SITC. Just tell us a little bit about what's been challenging about drugging STAT3 historically, and why you think a degrader
approach is well suited here.

DECEMBER 01, 2021 / 2:15PM, KYMR.OQ - Kymera Therapeutics Inc at Evercore ISI HealthCONx Virtual
Conference
Question: Maneka Mirchandaney - Evercore ISI - Analyst
: Are there any particular cancer types that you think are actually amenable to STAT3 degradation and which ones might those be?
Question: Maneka Mirchandaney - Evercore ISI - Analyst
: Awesome. I think that's all we've got the time for. I know there's so much else happening at Kymera. But, luckily, we've got the R&D Day coming
up in a couple of weeks to talk about some of that. But thank you, Nello, so much for joining. And thanks, everyone, for listening as well.
